Triclabendazole is a member of benzimidazole group of anthelmintics. It is a newly launched member with a very different spectrum of activity – directed against liver flukes. It is very potent against liver flukes (Fasciola hepatica) from 1 day old early immature to adult. Triclabendazole acts by interrupting parasite energy metabolism by binding to tubulin, thereby disrupting microtubular cell structure and preventing nutrient uptake and other functions. Triclabendazole acts primarily by binding to worms’ tubulin. Specifically it binds to dimeric tubulin, which, in turn, prevents the polymerization of tubulin during microtubule assembly. The earlier effects ascribed to triclabendazole, such as inhibition of fumarate reductase activity, may be secondary to the disruption of microtubules, which are essential for secretion of many enzymes.
